O'Dell graduated from the now-closed Coastal Academy High School in 1984. She went on to graduate Summa Cum Laude with a major in Marketing from Clemson University in 1990. During college she was a member of Kappa Kappa Gamma sorority (which included former SPEED Channel broadcaster Connie LeGrand). Then known as Nancy Humphries, she won the Miss South Carolina pageant in 1987. At Miss America, Nancy won a swimsuit preliminary in one of the toughest competitive years the pageant had ever seen.

In June 29, 2005, O'Dell married Keith Zubchevich, an executive for Riverbed Technology. They met while waiting in line at a Los Angeles-area airport.

On December 2006, she announced that she is expecting her first child in the summer of 2007.

She and Billy Bush currently serve as co-hosts of Access Hollywood, a 30-minute entertainment news program produced and syndicated by NBC Universal Domestic Television Distribution. She has been with the show since its 1996 inception, and became co-host in 1999. O'Dell serves as a contributor for NBC's Today Show and Dateline NBC. In addition, O'Dell co-hosted the Miss USA and Miss Universe competitions in 2004 and 2005, respectively. She also co-hosted the Miss USA 2006 pageant with Drew Lachey, the Miss Universe 2006 pageant with Carlos Ponce, as well as the recent Miss USA 2007 pageant with Tim Vincent.

O'Dell has appeared in several movies as either herself or a generic entertainment reporter, and has also taken on other acting roles, including minor roles in the television series Charmed (Season 1 finale and season 5 premiere). O'Dell was also the host and consulting producer of the USA Network talent competition Nashville Star over its first two seasons, but made the choice to leave the show upon its move from Saturday to Tuesday because of conflicts with her Access Hollywood schedule.
