Lee was born in São Paulo, Brazil, from an American father and an Italian mother. In 1966, together with Arnaldo Baptista and Sérgio Dias, she formed the band Os Mutantes. After she left the band in 1972, she has released many albums as a solo artist.

Rita Lee was married to her Mutantes fellow Arnaldo Baptista from 1971 to 1977. In 1996, she married Roberto de Carvalho, who was introduced to her by Ney Matogrosso.

Lee was educated in a French school, and speaks English, Portuguese, Spanish, French and Italian fluently.

Besides her musical career, she had a humor program (Radioamador) on radio in 1986, which aired for 9 months. She wrote 3 children books in 1986, and appeared on some Brazilian movies and TV shows. From 2002 to 2004, she was one of the hostesses of the Brazilian cable TV talk show Saia Justa. In 2005 she started her own talk show, called Madame Lee, along with her husband. Rita Lee is the namesake of Brazilian MPB singing star Maria Rita, daughter of friend Elis Regina.
